Well, I started a thread back in August (https://forums.joeuser.com/411269/) that's seen a lot of activity and actually had quite a few good bits of information for new and old players alike. I figure it's time to start a new thread and perhaps keep the OP up-to-date with useful information, etc. Feel free to use this post for any LoL discussion, etc.
Super fast background: I played a lot of Demigod as pacov/cheesuscrust. Back in August 2011 or so, I started getting heavily involved in LoL and folks have been kind enough to chime in with tips and links to various sites that have been quite useful to me. In addition, I've been able to keep up with folks that I've played Demigod with in the past and meet some new folks that play LoL and frequent these message boards.

k - so I'm just going to put down some of my thoughts. I'm not amazing at this game, but I do certain things that improve my odds of winning in ranked and so far its panning out just fine (bronze 5 to gold 5 in about a month or so). We all have varied skill levels, so some of this might be useful and some not. Caveat complete.
Prerequisite- you need to know how to play every single role. You don't have to be a god at every single role (more power to you if you are - I'm certainly not). You need to be able to play at a serviceable level in every single role and excel at least at 1 role (preferably 2). In my case, I'm good at support and adc. I'm not great, but have serviceable mids and jungles and so so tops. Knowing what you are good at and bad at is very important. If you don't know, I can probably tell you - but you really should know... Anyway, you need to have enough champs to make solid picks in any role and you need to be able to cover any role if it comes down to it. I'll talk about how to get better at specific roles in a bit.
Champ select advice - Don't be the "fill" guy unless you really feel like you are awesome in all roles. Call your preferred role immediately when you hit the lobby. Say "adc pref." Do this as soon as you hit the lobby. Some people believe that if you call a role you magically get it. This is stupid, but if you call something out, folks will often accomodate your request. If you are feeling wishy washy for whatever reason, call out multiple roles in order of what you want to play - "adc/mid pref." In my experience, you generally do not want to call support. It's a very important role, but you want to be in a position to carry every single game if possible. If you aren't calling out a role, you are hoping that other players can carry you. If you are hoping that folks will carry you, you don't deserve to win. You need to know your best roles and you absolutely should request them.
Solo or duo in ranked? - Well, my 1st thought here is that you should solo. If you solo, you have to rely on yourself. IMO - its a much bigger test of skill to play solo and win than to duo. Do you want someone to carry you or do you really want to know that you are actually good? That said, provided you have the right duo partner, you can increase your odds of winning quite a bit. The simple math is that instead of having 4 teammates as unknown quantities, you only have 3. If you have a good duo teammate, you know what you can expect. When you duo, you get the most bang for your buck by having complimentary roles. Support/adc is good; jungle/mid also works. Even if you don't have direct symmetry in roles, if you are an amazing mid and I'm an amazing adc, our odds go up quite a bit if we both get those roles. Our odds go way down if we can't get those roles and are forced to play out of positions that we are weaker. For example, Bryff is a good mid. If he duo's up with me, the only way he will get to play that role is for me to call mid, take it myself, and then put myself into a position like support where I can't carry out of (I'm always 1st pick, Bryff is always last pick when we duo)... that and I have to show the mid I'm picking at the start which means Bryff gets hard countered. In short, we can expect that we will be a disadvantage when duo queuing. Now, if Bryff queued up with someone at a lower elo than himself that was solid, he'd likely be 1st pick and be able to get mid and have a better chance of carrying the game. Anyway, you need to keep in mind where you end up in pick order when duoing if you aren't looking to go adc/support. If you want that, you can usually get it.
How do you get better in roles that you are not good at? - Well, here's what I do. And this is really what I do on a regular basis. I think about what I'm good at and what I'm not good at. For instance, I realized that I don't play any hyper carry ads. So, I read up on hyper carries like vayne and kog maw. I look for guides on how to build them (most adcs are the same btw ), I look for vids on how people play them (eg how do I all in with a specific champ - is there a combo - when do I all in - what's the best way for me to burst - how should I behave in lane with this champ). Then, I fire up a custom game and try out the mechanics of whatever champ vs AI. Usually I learn a few tricks during that custom on how to position myself, etc. Then, I'm off to normal games where I'll request the role or character I'm trying out. Now, people still report your for being awful in normals, but it really is where you need to try out characters to see if you are any good or not as the bots are useless for proving your skill level to yourself. So, fire up that normal and ask to play a role (again - after you've tried out the champ against bots). If you don't know anyone you are with and you are quite awful, I suggest muting everyone at the start of the game. Then, do your best. Continue with this until you feel like you have a serviceable skill level in whatever role. And keep in mind what you need to work on. For me, I noticed I didn't really have alot of mids for ranked, so I started practicing some with gragas against and karth. That way, I'll be able to get the job done if I need to play mid. The next thing I need to do is put more time into being better top lane. Again, because while I prefer adc/support, I might need to play top for the team. So, best to be ready for it. Put your time in and practice roles.
One last thought - there are certain things you can generally expect in ranked. 1 - if you are 1st pick, you generally can call whatever you want. Common knowledge, I'm sure - but I'll add - CALL WHATEVER YOU WANT. 2 - if you duo queue, you generally can lock down both adc and support. It works best if your adc is 1st pick, ofc. I've bumped into like 10 random dedicated supports in all of my games of LoL. People generally aren't going to call support. So, if you are 1st pick and duo'd, just call adc and then your duo partner can almost always get support.

Character guides
My nasus skill is unparalleled. If you watch, you know exactly how - god awful low skill cap late at night and with calculating the low dmg burst for last hitting. I'll like really, really weak mechanically with him. Sheen solves that problem and I typically go that on teams that have any sort of hard cc before I build a tank item (after of boots 2/upgraded jungle item)... that "somehow" fixes my mechanical problem.
Mouse of your Q to see how much damage it does, click on the monster to see how much health it has, only press Q when monster health is less than your Q damage. I had to do this when I used to play jungle Nasus because I was not a Nasus expert, but it meant that I had a lot more stacks than you did at the end of games. An extra 80 damage per Q matters at the end of the game.

@Pacov Do eeet! I know that it seems like MadCast is full of shiny happy people who just want to fool around all game, but I think there are more like minded folks who want to constantly improve out there than you think. Think of it as a call out to your kindred spirits.
@Cow Do eeet! They're trying to zero in on the best way to achieve some semblance of balance for the tournaments. The Halloween one I played in, they tried this thing where they ranked all the people in each role, and all the ADCs were captains. The worst ranked ADC got to pick his Mid player first and then they alternated through the roles that had all the people helpfully rated.
It went wrong in two ways. The one team with the best ADC ended up with three Diamond players and their two last pick players that didn't end up being that bad. In fact, their top did pretty damn well. Also, somehow I ended up as the best rated Jungler somehow. Or maybe I just got picked first because I was one of the better ones and they knew I'd camp the shit out of our Diamond I player's lane? Either way, they screwed up in that 3 First Picks/2 Last Picks > Any Other Picking Order and that Jungle didn't have a lot of strong players. I think they realize that and are going to adjust for the next one, but we'll see.
Either way, you and Karl should join up to play! I know that it secretly eats you up inside that I have a Gloriously Victorious Triumphantness Ryze skin that y'all do not.

Hrm, the big change to jungle items is going through next patch - Spirit Stone plus its upgrades get health/mana steal from minions (not just Spectral Wraith).
Thats pretty big. The easy availability of bigtime sustain from jungle very early from the obvious item path might open up new junglers.

My oh my did I not like that one chick that joined us for in house games last night. Kind of figured that would be the case - probably should have buggered off when I ended up on her team.
Anyway, I'm off tomorrow for my actual birthday... been a heck of a week what with the flu and early morning work. Just need to get through a full day of work today - which is going fine.
incidentally - if karl was a madcast candidate, he would have been invited to all of the in house games. if apoz is organizing, he typically tries to get candidates in for the in houses 1st.
So, I created yet another alt account that I started leveling on and off during my weird sleep schedule during flu sick times. I went ahead and dropped $10 on rp and picked up yasuo (riot gave me another 300-400 rp for hitting level 5). I've been playing yasuo only 100% of the time. Let me tell you of my super exciting adventures.
1 - NEVER, EVER, EVER play ARAM solo on on a low level smurf account. My queue time was like 8 minutes (they don't let you do ARAMs until you are like level 5 or so btw). Within the ARAM were exceptionally awful people on both teams doing nothing that made any level of sense. I had been doing bot matches up until that point. Anyway, it was just really, really, really, crazy. Like the first time folks have ever played a character crazy. Imagine 9 other people trying to figure out what abilities are all in a game mode they quite possibly have never played before... folks that don't even shop. It was a hot mess. Anyway, not going to do that again. Think the game was like an hour long...
2 - yasuo without runes/masteries = absolute rubbish early game. He starts getting towards god tier still once you get SS.
Other than that, I'm still rot with yasuo. I'm, ofc, doing quite fine against bots and in norms, but I'm not excellent with utilizing his w. Working on that a bit and still figuring out optimal placement. I'm getting quite good with using his dashes to stack damage and reposition intelligently, though. Not sure how far I'll go with the account, but I'm having fun here and there and not facerolling folks anyway.
one thing I don't know, but will test today at some point - do you get 3 stacks on yasuo's q if you do the e/q dash/blade thing and hit at a total of 3 minions. I believe that's how it works, but not sure. If so that will change my playstyle up some
Yeah, I'm not a fan of the whole "Let's have fun!" thing, cause usually the opposite occurs for me after someone on my team says that. Seems to me that people are under the impression that troll picks are more fun than they actually are, since their teammates are usually all playing something decent so the game doesn't end up completely lopsided. I wonder how many of those people would enjoy the 20 minutes of pure joy that games would be when everyone on their team was playing something non-viable?
Don't take it out on me next time though. We had a completely troll bot lane and a non-standard mid. Not sure what else you wanted? I've got a tourney to practice for and we weren't going to make that the poor Bronze III play some crappy champ with crappy summoner spells against a Gold III.
If you don't feel like bailing on those games, then next time fight trolling with trolling. Figure out a Jungle Kass build with 40% CDR and infinite mana, then only visit your allies lane to push the wave! Yell "Gank Tax!" when you do, even though you're not actually ganking anyone, cause you just play for fun. If you're getting trolled by a completely non-viable jungler like Teemo (or Kassadin bahahah!), just play Lee/Elise/Nid mid cause they're highly mobile and have execute abilities and take Smite. Then you can tell them how much fun you're having (for you personally, fuck the team) to have all the buffs all the time!

Well I am a sad cow. I valiantly tried to hit Diamond before they closed out the ranked season and made it to 95 LP in Plat I. I was almost certainly going to get it by the end of this week but SUPRISE today they are going to reset ranked queue. I was going to try to grind it out today but they disabled ranked queues at like 9am. OH well maybe this year....

well... just because they are turning off clamping doesn't mean they aren't going to make other LP adjustments.
Previously, the system could award more LP per win than it could take away. (LP not earned by "winning" while in a promo ~= LP not lost when dropping to 0 LP on the way down). They did this because it's more emotionally rewarding to gain 22 LP, but only lose 14 per loss. It feels like you are progressing and gradually gaining points. But, they could only do that because clamping in Division I existed. There wasn't a big risk of over-rewarding LP because if your MMR was too low and you got to Division I because of inflated LP gains... you would have to go through the trying process of a Division I promotion clamping.
Because clamping has been removed, LP gains per win are going to be similarily adjusted. i do still think that they will give slightly more LP per win than per loss (just because of promotion series "eating" some LP on the way up when you are in lower divisions). But i don't think diamond is going to suddenly be easier to get - espiecially since climbing is generally more difficult early in the season because of MMR deflation. (Eg, divisions are determined by the standard deviation of all players. Consistently thru the season, you have more new players going into ranked unprepared and "donating" some MMR into the system on their way to bronze. That MMR slowly drifted up the ladder and so it makes it easier and easier to get higher divisions as time goes on).

Previously they had a system that would clamp you hardcore when you got close to jumping up a tier, but wouldn't do much otherwise. I think that the big change is that the clamping is just going to be more spread out - instead of it being applied all at once when you get close to moving up, it will be applied in smaller amounts, but spread out over a larger range.
So if you add up all the clamping, it will be about the same as it was last season. They might not call it clamping any more because the shape of the adjustment is different, but the overall effect will be roughly the same.
It will just be less frustrating because you will move up in a more consistent and predictable way, rather than having stupid +1LP games. The behavior of LP gain when you got around 95LP in 1st division tiers really was bizarre and needed fixing.
The big question is when to play placement matches. I'm not sure if its smart to wait for the first wave of people to get placed or not.
